【关键词】 细胞凋亡 Effects of gossypol acetic acid on apoptosis and differentiation in leukemia HL60 cells 【Abstract】 AIM: To investigate the effects of gossypol acetic acid(GAA) on apoptosis and differentiation in classic human actue promylocytic leukemia cell line HL60 and study the possibility to treat leukemia with it. METHODS: The cytotoxic activity of GAA on HL60 cells was detected by means of MTT assay. Apoptosis was identified and analyzed with the help of transmission electron microscopy, cell cycle, and DNA gel electrophoresis, etc. The differentiation of HL60 cells into more mature granulocytes was detected by nitro blue tetrazolium (NBT) reduction test. RESULTS: More than 6 μmol/L GAA could suppress the proliferation and induce the apoptosis in HL60 cells in a dosedependent manner. NBT reduction test demonstrated that HL60 cells could also be induced to differentiate into more mature granulocytes by lowdose GAA (1, 4 μmol/L, P<0.05). CONCLUSION: GAA could selectively induce the apoptosis of leukemia HL60 cells in a smaller dose than that for human peripheral blood mononuclear cells, which suggests that it may be an ideal drug for leukemia therapy. 【Keywords】 gossypol; HL60 cells; cell division; apoptosis; cell differentiation 【摘要】 目的: 通过醋酸棉酚(GAA)对经典白血病细胞系HL60的研究,探索GAA治疗白血病的可能性. 方法: 用细胞培养、四唑盐比色试验(MTT)、细胞形态学、DNA电泳及NBT还原试验等方法观察不同浓度GAA促进HL60细胞凋亡及诱导分化的作用. 结果: GAA在浓度>6 μmol/L时可显著抑制HL60细胞的增殖,促进其凋亡,并表现出剂量效应;在诱导分化浓度下(1, 4 μmol/L),GAA可促进HL60细胞向成熟粒细胞分化(P<0.05). 结论: GAA对白血病细胞HL60的选择性促凋亡作用表明其可能是一种理想的高效低毒抗白血病药物. 【关键词】 棉酚;HL60细胞;细胞分裂;细胞凋亡;细胞分化 0引言 白血病发病率有显著升高的趋势[1]. 近年来,多酚类药物的抗肿瘤作用日益受到重视[2]. 作为多酚类药物的一分子,棉酚天然存在于锦葵科某些植物的根、茎、种子中,其化学结构为2, 2′联二萘8, 8′二羧基醛1, 1′, 6, 6′, 7, 7′六羟基5, 5′二异丙基3, 3′二甲基,分子质量为518.54,具有多种生物学活性[3]. 然而,以往曾经报道棉酚的抗肿瘤作用却未得到多数学者的认可. 近年随着研究的深入,其显著的抗肿瘤作用又重新得到了人们的重视[4]. 棉酚对白血病细胞的作用效果如何,有无临床使用价值?我们通过使用其衍生物醋酸棉酚(GAA)对经[1] [2] [3] [4] [5] 下一页 |
